Ganwyd John Penry yn sir Frycheiniog ac fe’i addysgwyd yng Nghaergrawnt. Ar y pryd roedd Elisabeth I ar orsedd Lloegr ac aeth Penri ati i feirniadu ‘r Eglwys Brotestannaidd a sefydlwyd ganddi hi a’i Senedd yn 1559. Daeth yn Biwritan ac ymuno â charfan y Presbyteriaid a dechrau ysgrifennu a chyhoeddi traethodau beirniadol ar gyflwr yr Eglwys. Aeth i’r Alban am gyfnod byr wedi iddo ymwneud â gwasg ddirgel yn Lloegr, ac yn 1593 fe’i cyhuddwyd o deyrnfradwriaeth yn y llys brenhinol yn Llundain ac fe’i crogwyd ym mis Mai y flwyddyn honno. Fe’i cydnabyddir yn ferthyr dros grefydd Biwritanaidd yn Lloegr a Chymru
